Effects of infusion of ergogenic acids on blood free fat acid and tryptophan hydroxylase (TPH) expression in rats

The pupose of this study was to investigate the effects of infusion of ergogenic acids on blood free fat acid (FFA) and tryptophan hydroxylase (TPH) expression in dorsal raphe. Male Sprague-Dawley rats were randomly divided into control (CON), exercise-with-NaCl (EXE), exercise-with-glucose (GLU), exercise-with-caffeine (CAF), exercise-with-carnitine (CAR) groups. Rats were injected intraperitoneally the glucose ( 1.4 g/kg), caffeine (6 ㎎/kg), carnitine (100 ㎎/kg), and 0.9 % NaCl 1 h prior to the exercise for 3 consecutive days . Animals of the exercise groups were put on running for 30 min once a day for 6 consecutive days. On the 7th day of the experiment, rats were run on a treadmill at a speed of 35 m/min until exhaustion. In the present results, exercise time to exhaustion in the CAF and CAR groups was significantly increased compared to the EXEl and GLU groups. Blood FFA levels in the CAF group was significantly increased compared to the EXE and GLU groups, and the CAR group was than higher the EXE group. TPH expression of the dorsal raphe in the EXE, GLU, CAF, CAR groups was significantly increased compared to the CON group. However, CAF group decreased TPH expression significantly compared to the group. These results suggest that caffeine, carnitine administration could promote fat oxidation, resulting in higher endurance performance during exercise, and especially the caffeine, inhibiting TPH expression, may play an ergogenic role in exercise performance by reducing fatigue.
