This study examined (1) whether a single session of exercise reduces the size of the myocardiac infarct after reperfusion. (2) whether exercise activates transcription and activation of iNOS for its upregulation. (3) whether exercise-induced late PC initiates induction of iNOS as meditater of late PC. In this investigation, iNOS knockout mice were used to role out the potential critical biases, because pharmacologic agents are not highly specific and do not entirely inhibit the target enzyme. The results were following: Exercise induced late preconditioning, having reduced infarct size from 72% to 31%. However, there were no differences of transcription and translation of iNOS, eNOS, and NF-kB between non-exercise and exercise including iNOS Knockout mice groups. These results suggested an alternative signaling pathway different from the late ischemic preconditioning. Therefore, there was possibility of another signaling pathway of preconditioning during exercise, and further studies are needed for the signaling pathway of exercise-induced preconditioning. |
