Effect of eccentric exercise on insulin resistance and skeletal muscle GLUT-4 protein expression of type Ⅱ diabetes rats
ABSTRACT
The purpose of this study was to investigate the insulin resistance and GLUT-4 protein expression in soleus and triceps according to downhill treadmill exercise in rats. 7-week-old, Wistar & GK rats were randomly divided into Non type 2 diabetes․Non exercise(NDM-NEX), Non type 2 diabetes․Exercise (NDM-EX), type 2 Diabetes․Non exercise(DM-NEX), type 2 Diabetes․Exercise(DM-EX). Rats in NDM-EX and DM-EX group were forced to run on he treadmill at the -16 degree and speed of 16m/min for 1 hour. All group rats were incubated with 2ml/kg glucose 20% solution after exercise and blood samples were taken from the tail at 0(before injection), 30, 60, 90 and 120 min after the oral glucose load for blood glucose. After 1 week wash-out period, rats in NDM-EX and DM-EX group were forced to run on he treadmill at the -16 degree and speed of 16m/min for 1 hour. After eccentric exercise in blood glucose, DM and NDM group showed high that compared before exercise to after exercise. In addition, compared with NDM-NEX group, NDM-EX group showed high insulin resistance during 120 min in insulin. On the other hand compared with DM-NEX group, DM-EX group showed high insulin resistance during 30 min and low insulin resistance after 30min in insulin resistance. In conclusion, the new finding of this study was that insulin resistance by down-hill treadmill exercise significantly different between DM group and NDM group.
Key words: eccentric exercise, insulin resistance, GLUT-4, type 2 diabetes
